Several limitations exist that can impair healthy development after birth (e.g. sheep Dolly) including telomere shortening problem. Briefly, the chromosome telomeres shorten after each DNA replication (in somatic cells) dues to the nature of lagging strand replication (Okazaki fragments). It is thought to be a contributing factor to aging, therefore the adult cell DNA that is transferred into an egg during reproductive cloning has already aged and contains shorter telomeres. The clone has therefore a potential to suffer from premature aging. The problem of replicating the ends of chromosomes is solved by a specialized enzyme (telomerase) that extends telomeres using an RNA template that is part of the telomerase. Telomerase enzyme is active during embryo formation (sexual reproduction) and in cancer cells.
